The present work was undertaken to study the effects of 24-epibrassinolide (0, 10-10, 10-8 and 10-6 M) on growth (shoot length and number of leaves), protein content and activities of antioxidative enzymes [guaiacol peroxidase (EC 1.11.1.7), catalase (EC 1.11.1.6), glutathione reductase (EC 1.6.4.2), ascorbate peroxidase (EC 1.11.1.11), superoxide dismutase (EC 1.15.1.1), monodehydroascorbate reductase (EC 1.1.5.4) and dehydroascorbate reductase (EC 1.8.5.1)] in leaves of 60 days-old Brassica juncea L. plants treated with different concentrations of Cr metal. In order to investigate the defense response in castor bean (Ricinus communis) against predators, we analyzed the effect of mechanical wounding upon the phospholipase A2 (PLA2) activity of leaf extracts. Time course experiments revealed that the highest levels of increased PLA2 activity (ca. two fold) occurred 15 min and 60 min after injury. The induced activities demonstrated high sensitivity towards aristolochic acid (10 mM), a PLA2 inhibitor. Based on SDS-PAGE analysis, the PLA2 activity induced 15 min after wounding migrated with a molecular mass of 40 kDa and was denoted RcPLA2 I. Potato tubers (Solanum tuberosum L. cv. Binje) were stored at 18 and 4ºC in order to verify changes in mitochondrial metabolism, especially in the activities of the alternative oxidase (AOX) and plant uncoupling mitochondria protein (PUMP). Tubers were also sliced and submitted to artificial aging. The oxygen consumption rate (OCR), respiratory control (RC) and ADP/O ratio for mitochondria obtained after storage at 18ºC were not altered, regardless of the time of storage. Considerable increment in OCR, for both respiratory states 3 and 4, was observed after storing the tubers for five and ten days at 4ºC; and it was accomplished by reductions in RC and ADP/O. Yellow vein mosaic disease of mesta (kenaf, Hibiscus cannabinus L.; and roselle, H. sabdariffa L.) is a new entrant to the disease scenario and it is associated with a novel monopartite Begomovirus. Changes in different biochemical parameters in diseased mesta plants were observed as compared to healthy ones. Isozyme pattern and assays of different enzymes, namely catalase, acid phosphatase, peroxidase, esterase, polyphenol oxidase and superoxide dismutase, revealed lower activities of catalase, acid phosphatase and peroxidase enzymes and enhanced activities of esterase, polyphenol oxidase and superoxide dismutase in diseased plants as compared to healthy ones. Three degrees of red mite infestation (Oligonychus ilicis McGregor) (Acari: Tetranychidae) were evaluated with regard to leaf anatomy, photosynthetic gas exchange and leaf ethylene production, in a commercial crop of approximately 12-year-old trees of Coffea arabica 'Catuaí Vermelho'. As a complementary study, foliar anatomy was also evaluated in a recuperated crop of C. arabica 'Mundo Novo' that had presented a high degree of red mite infestation during the dry period of the prior growing season. The transport of organic N compounds to the shoot in the xylem sap of nodulated soybean plants was investigated in an attempt to better understand the changes in N metabolism under root hypoxia (first 5 days of flooding), with different sources of N in the medium. NO3- is beneficial for tolerance of plants to waterlogging, whereas other N sources such as NH4+ and NH4NO3, are not. Nevertheless, in the presence of NH4+ high levels of amino acids were transported in the xylem, consistent with its assimilation. Some increase in the transport of amino acids was also seen with NO3- nutrition during waterlogging, but not with N-free medium. Rubber production is dependent on the availability of carbohydrates in the tissues engaged in latex synthesis. In this study three rubber tree clones were used to study the protein profile, the activities of invertase and sucrose synthase in the latex and anatomical characteristics of the bark, as well as to establish a possible relationship with latex production. The clones used in the study, RRIM 600, GT 1 and Fx2261, differed in latex productivity, from high to low in this order. The antioxidant defense system response was evaluated in two rice cultivars (Oryza sativa L.), Fernandes (CNA-1158) and Maravilha (CNA-6843-1), treated with toxic levels of aluminum. After exposure to aluminum we determined plant growth, H2O2 and O2•- contents, lipid peroxidation, antioxidant enzymes activities and ascorbate and dehydroascorbate contents. Al predominantly accumulated in roots of both cultivars but it reduced root and shoot growth only in the Maravilha cultivar. Treatment with aluminum resulted in a reduction of 84 and 60% in the levels of H2O2 in the roots of the cultivars Maravilha and Fernandes, respectively, and of 26% in the levels of O2•- only in the roots of Fernandes cultivar.
