Activation of nitrate reductase of cashew leaf by exogenous nitrite Braz. J. Plant Physiol.

Nitrate reductase (NR) plays a central role in plant primary metabolism and exhibits complex regulation mechanisms for its catalytic activity. There is controversy in the literature concerning the possible direct effect of NO2- on the stimulation and/or activation of NR activity. The influence of NO2- was studied on the NR activity in the leaves of 30-day-old cashew (Anacardium occidentale L.) seedlings. Addition of NO2- to the reaction mixture containing leaf enzymatic extract resulted in a remarkable increase in NR activity. Activation of phenylalanine ammonia lyase as a key component of the antioxidative system of salt-challenged maize leaves Braz. J. Plant Physiol.

Differential antioxidative activities were assessed in the leaves of two maize inbreds (A-180 and A-619) under salt stress and the subsequent recovery period. Total antioxidation test revealed that in both inbreds, this ability was sharply increased during stress period, but was slowly reverted back to the normal level during recovery. The enzymatic antioxidative analysis showed differential patterns in the activities of catalase, peroxidase and polyphenol oxidase in both maize inbreeds. Activation of phospholipase PLA2 actvity in Ricinus communis leaves in response to mechanical wounding Braz. J. Plant Physiol.

In order to investigate the defense response in castor bean (Ricinus communis) against predators, we analyzed the effect of mechanical wounding upon the phospholipase A2 (PLA2) activity of leaf extracts. Time course experiments revealed that the highest levels of increased PLA2 activity (ca. two fold) occurred 15 min and 60 min after injury. The induced activities demonstrated high sensitivity towards aristolochic acid (10 mM), a PLA2 inhibitor. Based on SDS-PAGE analysis, the PLA2 activity induced 15 min after wounding migrated with a molecular mass of 40 kDa and was denoted RcPLA2 I. Activity of alternative oxidase and plant uncoupling mitochondrial protein in potato tubers stored at low temperature or submitted to artificial aging Braz. J. Plant Physiol.

Potato tubers (Solanum tuberosum L. cv. Binje) were stored at 18 and 4ºC in order to verify changes in mitochondrial metabolism, especially in the activities of the alternative oxidase (AOX) and plant uncoupling mitochondria protein (PUMP). Tubers were also sliced and submitted to artificial aging. The oxygen consumption rate (OCR), respiratory control (RC) and ADP/O ratio for mitochondria obtained after storage at 18ºC were not altered, regardless of the time of storage. Considerable increment in OCR, for both respiratory states 3 and 4, was observed after storing the tubers for five and ten days at 4ºC; and it was accomplished by reductions in RC and ADP/O. Yellow vein mosaic disease of mesta (kenaf, Hibiscus cannabinus L.; and roselle, H. sabdariffa L.) is a new entrant to the disease scenario and it is associated with a novel monopartite Begomovirus. Changes in different biochemical parameters in diseased mesta plants were observed as compared to healthy ones. Rice is a tropical plant, so cold temperature may be detrimental to its development, depending on the genotype and environmental conditions. Degree of lipid unsaturation has been related to cold tolerance due to its effect on membrane stability. So, the aim of this study was to characterize the fatty acid composition and its alterations due to cold temperature in rice genotypes of diversified origin. Forty-four rice genotypes at the V4 stage were submitted to two temperature conditions: 10°C and 28°C for two days and after this they had their leaves collected for lipid extraction and quantification. Three degrees of red mite infestation (Oligonychus ilicis McGregor) (Acari: Tetranychidae) were evaluated with regard to leaf anatomy, photosynthetic gas exchange and leaf ethylene production, in a commercial crop of approximately 12-year-old trees of Coffea arabica 'Catuaí Vermelho'. As a complementary study, foliar anatomy was also evaluated in a recuperated crop of C. arabica 'Mundo Novo' that had presented a high degree of red mite infestation during the dry period of the prior growing season. Ameliorative effect of exogenous nitric oxide on oxidative metabolism in NaCl treated chickpea plants Braz. J. Plant Physiol.

Accumulation of salts in irrigated soils is one of the primary factors limiting yield in South Asia. We investigated whether exogenous nitric oxide (NO) supplementation as sodium nitroprusside has any ameliorating effect against NaCl induced oxidative damage in chickpea leaves. NaCl treatment (250 mM) alone and in combination with two concentrations of SNP (0.2 and 1 mM) were given to 50 days old chickpea plants for 2, 4 and 6 days. Salt stress adversely affected the relative membrane injury, lipid peroxidation levels, relative water content (RWC) and H2O2 content. The effect was time dependent. Amino acid and ureide transport in the xylem of symbiotic soybean plants during short-term flooding of the root system in the presence of different sources of nitrogen Braz. J. Plant Physiol.

The transport of organic N compounds to the shoot in the xylem sap of nodulated soybean plants was investigated in an attempt to better understand the changes in N metabolism under root hypoxia (first 5 days of flooding), with different sources of N in the medium. NO3- is beneficial for tolerance of plants to waterlogging, whereas other N sources such as NH4+ and NH4NO3, are not. Nevertheless, in the presence of NH4+ high levels of amino acids were transported in the xylem, consistent with its assimilation. This paper describes a new instrumental methodology proposed for the study of seed germination which can be applied to germination physiology, agriculture and forest restoration. This instrument performs the recording of images of germinating seeds through the use of a flat-bed scanner coupled to a computer, where a specific program commands the capture of these images at predetermined time intervals. These images are archived for posterior analysis. The device allows the collection of data and the elaboration of germination curves with high accuracy. Rubber production is dependent on the availability of carbohydrates in the tissues engaged in latex synthesis. In this study three rubber tree clones were used to study the protein profile, the activities of invertase and sucrose synthase in the latex and anatomical characteristics of the bark, as well as to establish a possible relationship with latex production. The clones used in the study, RRIM 600, GT 1 and Fx2261, differed in latex productivity, from high to low in this order. High temperature and salinity are the major ecological factors challenging crop productivity in the arid and semiarid regions of the world. Effects of high temperature (43-45°C) and salt stress (0.6 M) on Macrotyloma uniflorum (Lam.) Verdc. (Horse gram), were evaluated in terms of antioxidants and antioxidant enzymes. Both treatments caused typical stress responses in this tropical leguminosae. Oxidative stress indicators such as H2O2, TBARS, and proline were significantly elevated. Antioxidant system response induced by aluminum in two rice cultivars Braz. J. Plant Physiol.

The antioxidant defense system response was evaluated in two rice cultivars (Oryza sativa L.), Fernandes (CNA-1158) and Maravilha (CNA-6843-1), treated with toxic levels of aluminum. After exposure to aluminum we determined plant growth, H2O2 and O2•- contents, lipid peroxidation, antioxidant enzymes activities and ascorbate and dehydroascorbate contents. Al predominantly accumulated in roots of both cultivars but it reduced root and shoot growth only in the Maravilha cultivar. Antioxidant-enzymatic system of two sorghum genotypes differing in salt tolerance Braz. J. Plant Physiol.

Two forage sorghum genotypes were studied: CSF18 (salt-sensitive) and CSF20 (salt-tolerant). Shoot growth reduction as a result of salt stress was stronger in the salt sensitive genotype compared to the salt tolerant one. When the two genotypes were subjected to salt stress (75 mM NaCl) no significant change in lipid peroxidation was observed. However, salt stress induced increases in superoxide dismutase and catalase activities in both genotypes. These salt-induced increases were higher in the salt-tolerant genotype.
